赤外円二色法によるスタンダード・キロプティカル構造解析法の構築

Research Abstract

昨年に引き続き、糖脂質合成をおこなった。保護糖によるグリコシル化反応の条件を検討し、グリコシル化に成功した。S及びR-コンフィグレーションを有するグリセロール、α及びβ体、グルコース及びガラクトース誘導体、合成8種類の糖脂質の合成を完了した。合成した糖脂質およびジアシルグリセロールのVCD測定を行った。VCD測定の際、測定温度、溶媒等の条件の検討を綿密に行い、最適条件を発見した。ジアシルグリセロールにおけるVCDにおけるカルボニルの領域のシグナルは、分裂型様の対称性のよい吸収であり、絶対配置判別に特徴的であることが判明した。本シグナルは、旋光度では識別不可能なジアシルグリセロールの絶対配置決定にVCDが極めて有効であり、唯一の非破壊的方法になりうることを示している。また、糖脂質においては、同様なカルボニル領域のVCDシグナルに顕著な差が観測され、一般的な経験則の確立が可能であることが判明した。上記で得られたVCDスペクトルに関する理論計算を試みた。脂肪酸の側鎖部分が短い(C4 or C8)ジアシルグリセロールに関しての理論計算を開始した。従来行っているConflex社製のプログラムとGaussaian 03の併用をメインとして、実測VCDスペクトルとの比較により、基底関数などのパラメーターの最適化を行った。トリアシルグリセロールは、脂肪酸が3ヶ結合する脂質である。結合位置によるキラリティーの違いを考慮して、まず、最初に1位、2位に同一の脂肪酸を有するトリアシルグリセロールの合成を行い、2種の絶対配置が確定した純粋なトリアシルグリセロールの合成に成功した。
