Research Abstract

高速液体キャピラリークロマトグラフィー(HPLC)モノリス型シリカカラムの調製反応液の組成、すなわちPEG濃度と,テトラメトキシシラン(TMOS)-メチルトリメトキシシラン(MTMS)の濃度比を変化させて、高速、高性能分離に適した組成、濃度を検討した。
(1) TMOS:MTMS=3:1において調製した3-5m長のカラムは高い透過率(5μm粒子の約5倍)と約7μmの理論段高を示し、2-3本連結して7-12m長とし使用した場合、通過時間(t_0)=1.5時間で80-100万段以上の理論段数を発生して、スチレン5量体8種のジアステレオマーの完全分離や、1個の重水素置換によるベンゼン同位体化合物の分離を可能とした。また、3mと30cmのカラムをBSA消化ペプチドの分離に使用した場合、同一グラジエント勾配において長いカラムが3-5倍のピークキャシティを与えた。しかし試料の希釈により低いプロテインスコアやペプチドカバレージが得られた。
(2) TMOS:MTMS=75:25、85:15、90:10においてシリカ量を従来より約40%増加させて調製したシリカは優れた高速性能を示し、5μm粒子充填カラムの同程度の透過率と、2.5μm粒子相当の理論段高(5.5μm)を示した。これらのシリカは従来の組成で調製されたシリカより高い構造均一性を示した。
さらに均一な構造により、高速性能あるいは高理論段数をさらに向上させることが可能であると考えられる。また低濃度試料のために大量注入法の検討が必要である。
