尿路病原性大腸菌が産生するUSPの作用解析と粘膜ワクチンの開発

Research Abstract

1アミノ酸置換無毒化USP融合蛋白(mUSP-GST)を高純度に精製して抗mUSP-GST家兎血清を調整し、この抗血清を更にmUSP-GSTを結合させたAffinityカラムで精製してmUSP-GST特異IgGを作製した。この特異IgGを用いたSandwich-ELISA系を構築して精製mUSP-GSTに対する検出感度を検討したところ、検出限界は500pg/mlであった(論文準備中)。現在、この検出系を用いてUPEC臨床分離株のUSP産生量の調査とIntactなUSPの精製を行っている。
下部尿路感染患者に対して行う抗生物質の経口投与方法の基準化を検討した。102名の女性患者にgatifloxacinを1日当たり200mg、3日連続投与し、5-9日後に尿検査と問診を行い、4-6週後に症状の有無に関するアンケート調査を実施した。93.1%(95/102)の患者が治癒し、このうち感受性菌が原因の患者の治癒率は94.7%(89/94)、耐性菌が原因の患者の治癒率は75%(6/8)であった。4-6週後のアンケート調査では、55.9%(57/102)の患者から回答がよせられ、10.5%(6/57)の患者から症状の訴えがあった。今回の結果から、フルオロキノロンを1日1回、3日連続で経口投与する治療法は単純性膀胱炎患者の治療には効果的であると考えられる(Yamamoto S et al, J.Infect.
