In vivoパッチクランプ法を用いた下行性痛覚抑制系可塑的変化の解析

Research Abstract

脊髄痛覚シナプス伝達に対する脳幹からの下行性抑制作用を解析するために、ウレタン麻酔下に小脳を吸引・除去した後、刺激電極を定位に下部脳幹青斑核に留置し、脊髄後角表層、膠様質細胞からin vivoパッチクランプ記録を行った。
電流固定下に膠様質細胞から膜電位を記録すると、-70mV付近に静止電位を有し、自発性の興奮性シナプス後電位(EPSP)が誘起された。青斑核を電気刺激すると、EPSPの発生頻度や振幅に変化が見られたが、緩徐な膜電位の変化は観察されなかった。また、-70mVの電位固定下で青斑核刺激を行っても同様に、緩徐なシナプス電流は発生しなかった。次に、0mVの電位固定下で記録を行うと、膠様質細胞は自発性の抑制性シナプス後電流(IPSC)を誘起した。青斑核刺激を行うと、IPSCの発生頻度と振幅が著明に増大し、そのIPSCの増強作用は青斑核の電気刺激の強度に依存した。脊髄表面からノルアドレナリンを直接投与すると、予想外に膠様質細胞は電流固定下に10mV程度過分極した。-70mVの電位固定下ではノルアドレナリンによる緩徐な外向き電流が発生した。この外向き電流の逆転電位はK^+イオンの平衡電位である-90mVと一致し、電極内液に加えたCs^+やGDP-β-Sにより抑制された。また、0mVでは、ノルアドレナリンによるIPSCの発生頻度と振幅の著明な増大が、青斑核刺激の時と同様に観察された。脊髄スライス標本でも同様に、ノルアドレナリンは膠様質細胞に外向き電流を発生させる事、IPSCの発生頻度を増大する事が観察されている。以上より、脊髄表面に投与したノルアドレナリンの作用とは異なりin vivoでは、青斑核刺激により膠様質細胞に緩徐なシナプス応答や過分極は発生しない、一方で、抑制性のシナプス伝達を増強して痛覚伝達を抑制する事が示唆された。
