Research Abstract

細胞分裂の正しい進行のために特定の細胞周期で不要なタンパク質を処理するシステムが細胞内には存在する。その代表的分解コンパートメントであるプロテアソームの標的となるタンパク質はユビキチン鎖というマークが付けられる。
このユビキチン付加反応は、ユビキチン活性化酵素(E1)→ユビキチン結合酵素(E2)→ユビキチンリガーゼ(E3)の三段階の酵素反応で行われる。本課題では、このE3に相当する巨大なタンパク質複合体であるAPC/サイクロソーム(APC/C)の機能制御に関与するタンパク質分子の構造とはたらきに着目し、
(1)APC/Cの阻害因子を用いた解析
(2)APC/Cの活性部位構成分子の解析
(3)APC/Cの全体構造についての解析
を計画した。
