Outline of Final Research Achievements

The aim of this study was to develop glomerular and tubular models by 2- or 3-dimensional co-culture of human kidney-derived cells in a microfluidic device. Using the fabricated device, we optimized the culture conditions for kidney-derived cells by investigating different culture conditions. In addition, the characteristics of the models were clarified for the model in which shear stress due to medium flow was applied and for the disease model constructed by applying disease-inducing factors to the normal model.

Academic Significance and Societal Importance of the Research Achievements

開発した腎モデルを用いることにより、将来的には正常な腎排泄機能を維持するために必要な因子や、疾患を引き起こす因子、治療薬の探索が可能になり、腎臓病の治療法の開発や創薬につながることが期待される。さらに、本研究で検討した剪断応力下での共培養技術を応用することにより、将来的には他の臓器・組織の血管や消化管など、生体内のさまざまな管腔における物質透過性試験やその疾病モデルへの幅広い応用が可能となり、医学薬学への幅広い波及効果が期待される。
