Outline of Annual Research Achievements

これまで代表者らは、採取が容易な糞から性ホルモンを分析し、多くの希少種の繁殖生理を明らかにしてきた。
本研究では、希少ネコ科動物に着目し、①妊娠時にのみ検出されるホルモンを同定し、その濃度測定による繁殖率推定法を確立、および②これまで報告されていないネコ科動物の糞DNAメチル化検出による年齢推定法の確立を目指した。
2021年度は、昨年度に引き続き、動物園などの飼育個体の血液・糞により、妊娠・年齢推定法の確立を目指した。なお、サンプリングに関しては、飼育個体に加えて、海外協力機関により野生個体の採糞を実施し、カメラトラップによる餌動物密度調査、アンケートによる密猟・報復殺数の調査も実施した。
繁殖率推定法の確立に関しては、飼育個体の糞を用いて、高速液体クロマトグラフィー・質量分析計(LC-MS)などの分析手法により、糞中の性ホルモンの同定を試みた。また、糞から簡易的にステロイドホルモンを抽出し測定するField-friendly法を考案し、その概要を英語論文としてまとめ、国際誌で発表した。
年齢推定法の確立に関しては、年齢が判明しているイエネコおよび動物園や保護センターなどで飼育されている個体の血液からDNAを抽出し、バイサルファイト処理の後、PCR増幅、シーケンスした。バイサルファイト非処理試料と塩基配列を比較して、メチル化の程度を検出した。哺乳類(マウスやラット)で年齢によるメチル化の増加や減少が報告されている遺伝子ZC3H12D、BCAS4、cg063794355)を候補遺伝子と比較した。本研究成果は国際誌に投稿し、英語論文として発表した。

Reason

昨年度に続き、新型コロナウイルス感染症の拡大により海外渡航などに制限がかかったが、国内の動物病院や動物園、保護センターの協力の下、すでに採取済みであった血液や糞サンプルを提供いただき研究に供することで、当初の研究計画を完了するに至った。

Strategy for Future Research Activity

血液サンプルで得られた結果を、夾雑物の多い糞に応用して、年齢推定および妊娠診断法を確立する。
動物病院や動物園などの飼育個体だけでなく、野生下で採取した糞にも応用する。
