Outline of Annual Research Achievements

我々のグループは放射線と併用することで大量の活性酸素を産生し、放射線増感効果を著明に増加させる過酸化チタンナノ粒子の開発に成功し、この過酸化チタンナノ粒子は、良好に腫瘍細胞にエンドサイトーシスで取り込まれ、X 線照射によりヒドロキシルラジカルを始めとする大量のROS を発生させ、さらに殺細胞効果に繋がるなどを発見し、過酸化チタンナノ粒子は新たな放射線増感剤として米国、欧州、中国等、世界5か国以上で国際特許を得ており、世界的に先駆的な立場で過酸化チタンナノ粒子を利用した放射線増感剤の開発に取り組んでいる。腫瘍局所において過酸化チタンナノ粒子がX 線照射により大量のROS を発生し強い抗腫瘍効果を発現することがすでに確認されているが、腫瘍浸潤リンパ球(TIL)やPDL1発現の増加し、免疫チェックポイント阻害剤(ICI)を併用することで、さらなる局所的な抗腫瘍効果が高まる結果を得た。この過酸化チタンナノ粒子に関しては、マウス悪性黒色腫などの腫瘍モデルを用いた検討では、免疫療法との併用により、極めて優れた腫瘍増殖抑制効果を発見するなど複数の研究結果を得ている。この免疫サイクルの増強はアブスコパル効果をも増強し、放射線照射をしていない部位の腫瘍にも著明な細胞増殖抑制効果を確認している。この機序は、放射線照射で免疫原性細胞死が起こり、多彩ながん抗原の放出(Eat me signal)と樹状細胞からTILへの抗原提示の伝達が行われ、大量のTILが腫瘍へと浸潤し免疫サイクルが加速した。過酸化チタンナノ粒子を併用し小胞体ストレスを誘導しMHC class I の発現の増強を実現でき、免疫チェックポイント阻害剤の併用により、圧倒的な免疫原性の獲得に成功し、世界的にも類を見ない併用効果をマウス悪性黒色腫モデルで実現し、その成果は更なる国際特許申請に至った。
