慢性ヒ素中毒による肺がん発生に特異的なmiRNAを標的とした新規予防法の開発

Outline of Annual Research Achievements

ヒ素の中毒について、急性中毒には金属キレート剤が限られた効果を示す以外に特効薬がない。慢性ヒ素中毒による発がんにも予防治療薬がないのが現状である。本年度は、最も毒性が高い有機ヒ素化合物の一つであるフェニルアルシンオキシドの曝露における予防治療対策として、フェニルアルシンオキシド急性曝露から細胞死を抑制する天然薬物を調べた。フェニルアルシンオキシドはビシナルシステインとジスルフィドを形成し、細胞内活性酸素の生成を増強することで、活性酸素の生成と細胞内抗酸化システムの変化に注目して研究を進めた。ヒト表皮角化細胞株(HaCaT 細胞)を用いて、バイカリン(ブルースカルキャップなどのタツナミソウ属植物の種から抽出した成分)の存在下で、細胞培養液にフェニルアルシンオキシドを添加し、さらに24時間培養した。その結果、フェニルアルシンオキシドの曝露により細胞の生存率が顕著に低下し、カスパーぜ-3の活性化に伴いアポトーシスの誘導が検出された。バイカリン併用処理はその濃度に依存して、HaCaT 細胞の生存率を顕著に回復させ、アポトーシスの誘導を有意に抑制した。また、抗酸化作用を持つタンパク質の発現変化を調べた結果、バイカリン併用処理の防護作用に連動して特異的な発現変化を示す抗酸化酵素タンパク質が同定され、フェニルアルシンオキシドの曝露におけるバイカリンの防護作用には細胞内酸化ストレスシグナル伝達系の変化が寄与することが判明した。

Reason

バイカリンにより、フェニルアルシンオキシドの曝露が誘導する細胞生存率の低下が回復され、細胞死が抑制されることを見出した。さらに、その防護作用に寄与する抗酸化作用を持つ酵素タンパク質を同定した。

Strategy for Future Research Activity

フェニルアルシンオキシドの曝露による細胞生存率の低下、アポトーシスの誘導及び抗酸化酵素タンパク質の発現抑制におけるバイカリンの防護効果についてその詳細な分子メカニズムを解析する予定である。
