Outline of Annual Research Achievements

申請者らは、ヒトの鉛の警告濃度である5μg/dLに対して、トカゲで最大値 1875 μg/dL(警告濃度の375倍)、野生ラットで最大値 592 μg/dL(警告濃度の約120倍)という高濃度の鉛が検出されたにも関わらず、血液・臓器毒性などを示さない耐性個体群を見出した。本研究の目的は、高精度XAFS解析を多階層の動物種に応用し、これまで全く報告のない動物種普遍の共通の金属耐性メカニズムを解明することである。ザンビア共和国で採取した生体試料(野生ラット、ニワトリ、野生トカゲ、イヌの臓器など)と環境試料(土壌、水など)を対象とし、世界で初めて陸生野生動物における高精度X線吸収分光法(XAFS)解析を行うことを当該年度の目標としている。フォトンファクトリーにおいて既に野生ラットおよびニワトリにおけるXAFS解析を実施しデータを取得済みである。これらのデータから、哺乳類と鳥類の類似性・共通性のある結果も観察され、統計学的な解析を実施している段階である。また野生トカゲおよびイヌの全身臓器を採材し、その金属濃度の測定を行っている。高濃度を示す個体や臓器をもとにXAFS解析に使用するサンプルを選定している。また、鉛の化学形態が体内動態や蓄積特性に及ぼす影響を明らかにするために、マウスを用いたIn Vivoでの投与実験も開始しており、今後に得られるデータをもとに異なる形態の鉛を暴露した各種動物における血液毒性、臓器毒性を精緻に検査する。

予定通りにXAFS解析を行いデータを取得しているため計画通りに進展していると判断した。

Strategy for Future Research Activity

引き続きXAFS解析を進めていく。また、特に高濃度の鉛が検出された腎臓および肝臓においては、臓器ホモジネートの超遠心画分におけるLiquid Chromatography ICP-MS(LC-ICP-MS)などの組み合わせによる結合タンパク質の探索および、候補タンパク質に関しては、LC/MS/MS法によるタンパク質の同定を行う。

COVID-19により想定よりも出張が少なくなったなどの理由のために支出額が減ったため。
