Outline of Annual Research Achievements |
野生ラットの臓器および内容物の鉛化学形態分布について、Pb L3端 XANESスペクトルによる解析を進めた。高濃度の鉛で汚染されたラットであることに加えて、凍結乾燥による試料均一性の増大や鉛濃度の濃縮によって、これまで鉛低濃度により測定が制約されていた生物試料のXANESスペクトルの臓器別、内容物別の測定に成功した。野生ラット3個体を主として、臓器は心臓、脳、肺、脾臓、肝臓、腎臓、筋肉など、胃・小腸・大腸の内容物、および糞便に対して、スペクトルの質を考慮したうえで系統的な解析を試みた。鉛化学形態の参照用XANESとして、グルタチオンと鉛の結合体や経口曝露源と考えられる現地の鉛汚染土壌等のデータを用いた。スペクトル形状および参照スペクトルとの線形結合フィッティングの結果、臓器別の鉛形態の違いや、消化器系や体内へ吸収された際の鉛化学形態の変化に関する知見を得ることができた。トカゲ亜目(Trachylepis wahlbergii)計112匹の臓器(肝臓、肺、卵巣/精巣)、血液、胃内容物を採集し肝臓中の金属濃度の測定を行った。土壌、トカゲ臓器、生体内で鉛と結合することが知られているグルタチオンなどと鉛の結合試料のPb L3端 XANESスペクトルによる解析を行った。鉛の周囲にグルタチオンをはじめ硫黄化合物が存在することが示唆された。このため、硫黄K端のエネルギー領域も計測し、個体間及び臓器間で異なるスペクトルパターンを得た。化学形態に加えて多角的なデータの取得を目的に遺伝子解析も実施しデータベース上にない全ゲノム配列データを得た。肝臓、肺、卵巣ついてRNA-seqも実施した。関係省庁・現地獣医師の協力の下、ニワトリ(10個体)、イヌ(26個体)の死亡個体の肝臓、腎臓、骨、腸管、脳、心臓、脾臓を採材し、鉛など10元素の濃度測定も行った。ラットの動物実験による確認実験も実施している。

Strategy for Future Research Activity |
今後は、今年度に引き続きXAFS解析、LA-ICP-MS解析などを行う予定である。遺伝子解析やエピジェネティクス解析も同じ個体から行うことで、遺伝子変異以外の金属耐性機構のメカニズム解明を行っていく予定である。

Causes of Carryover |
コロナウイルス感染症により予定していた海外渡航およびこれに関わるサンプリングの謝金支出が大幅に減額されたため。次年度には予定通り海外渡航を行う予定である。
