網羅的ゲノム・エピゲノム解析による小児固形腫瘍の分子機構の解析と分子標的療法の開発

Research Abstract

我々はAffymetrix社のSNPアレイを用いて神経芽腫200例を検索し、MYCN(2p24)より少しcentromere側の2p23領域で増幅領域をみい出し、これがALK遺伝子であることを同定し、さらにALK遺伝子の活性化変異が神経芽腫の進展に関与することをみいだした(Nature,2008)。今年度はALKの発現解析の結果、NB細胞株NB-1ではexon 2、3を欠失した部分欠失型転写産物を発現していることを見出した。NB-1のこの部分欠損型転写産物はゲノム上のintron 1から4の約300kbのゲノムの欠失により生じていることが判明し、またアレイ解析によりこの欠失アレルが有意に増幅していることが見出された。部分欠失型ALKは強い自己リン酸化能力をもち、NIH3T3細胞を使ったcolony形性能やマウスの移植実験でも強い造腫瘍性を示した。また、ALK阻害剤により部分欠損型ALKを発現するNB-1では野生型ALKを発現する細胞に比べて著しい増殖抑制が観察され、部分欠損型ALKの活性化がNB-1の悪性化に重要であると考えられた。部分欠損型ALKも変異、転座、増幅の異常と同様に活性化をきたし、腫瘍化に関与することが示された。Ewing/PNET family(ESFT)97検体(細胞株17株、新鮮腫瘍80例)でRT-PCRによる発現解析、DHPLC法、直接塩基配列決定法を用いた変異解析、SNP arrayによるALK領域のゲノムコピー数の解析、NIH3T3細胞発現系を用いて、ESFTで検出された変異ALKの機能解析を行った。ALKの発現はRT-PCRを施行したESFT臨床検体46例全例で確認された。変異解析により、ESFTの中では極めて稀な鼻腔原発の2例にkinase domainの胚細胞変異を検出し、活性型変異であることを確認した。
