微生物の生産するアルベカシン耐性克服活性物質の探索

Research Abstract

近年問題になっているアルベカシン耐性MRSAはアミノグリコシド修飾酵素が耐性化に関与しているため、その阻害剤が得られれば耐性が克服できると考えられる。そこでアルベカシンと共存しているときにMRSAの阻止円径が大きくなる物質のスクリーニング系を利用して、アルベカシン耐性克服物質のスクリーニングを微生物培養液より行った。前年度は糸状菌Penicillium sp.FKI-4429の生産する新規物質biverlactones A-Dの単離を報告したが、今年度は新たに糸状菌Simplicillium sp.FKI-5985の培養液中より逆相シリカゲルカラムクロマトグラフィーやHPLCを用いて精製を行い、新規物質aogacillins A&Bを単離することができた。Aogacillin類はNMRなど各種機器分析を行った結果、エキソメチレンの結合したスピロ-δ-ラクトン-5-シクロヘキサン骨格を有するユニークな構造であることが決定できた。またaogacillin AとBはいずれも0.5μg/ml(この濃度ではMRSAに対する抗菌活性を示さない)をアルベカシンと併用することで、アルベカシン耐性MRSAに対するアルベカシンのMICを32倍増強する優れたアルベカシン耐性克服活性を示した。次年度は、biverlactone類およびaogacillin類の作用機構や動物実験における治療効果の検討を中心に、研究を進める予定である。
