トキソプラズマ原虫における植物ホルモン生合成能の存在意義の解明

アブシジン酸阻害剤以外の他の植物ホルモン生合成阻害剤やその他の植物生理活性阻害剤(除草剤など)をトキソプラズマ原虫やマラリア原虫の培養に添加してその影響を調べてみたところ、いくつかの物質でトキソプラズマやマラリア原虫の増殖を阻害していることが明らかとなった(論文投稿中)。そこで、そのうちの一つであるサイトカイニンに着目して、より詳細な検討を加えた。その結果、サイトカイニンはトキソプラズマのある種のサイクリンの発現量を調節しており、その結果原虫の細胞周期を制御していることが明らかとなった。さらにこの細胞周期制御により、サイトカイニンは原虫にとって必須の細胞内小器官であるアピコプラストの分裂をも制御していることを見出した。(投稿準備中)。
また、植物において報告のあったUPLC-ESI-qMS/MS法による植物ホルモンの高感度・大量解析法を原虫サンプルに応用できる可能性を検討した。その結果本法は原虫においても応用可能であることが判明した。現在本法を用いてトキソプラズマがどのような植物ホルモンを産生しているのか、またそれらの中間体を検出することによる生合成経路の同定を行っている。
