2021 Fiscal Year Research-status Report

Protein import into endosymbionts becoming organelles

Research Project

Project/Area Number 21K15094
Research InstitutionOkinawa Institute of Science and Technology Graduate University

Principal Investigator

HUSNIK Filip  沖縄科学技術大学院大学, 進化・細胞・共生の生物学ユニット, 准教授 (30886130)

Project Period (FY) 2021-04-01 – 2023-03-31
Keywordsproteomics / metabolomics / symbiosis / bacteria / insects / immunohistochemistry / 3D imaging / protein import
Outline of Annual Research Achievements

Metabolomics and proteomics protocols were established for the model system. All tissues of interest were prepared, their composition analyzed with mass spectrometry, and numerous metabolites identified. Volume imaging(microCT, FIB-SEM, confocal z-stacks) was used to understand host-symbiont integration. Protein candidates for future antibody labeling were identified.

Current Status of Research Progress
Current Status of Research Progress

2: Research has progressed on the whole more than it was originally planned.

Reason

Proteomics and metabolomics progressing more smoothly than expected. More imaging methods than originally planned were established, but antibody labeling has not been carried out yet.

Strategy for Future Research Activity

I will build on proteomics and metabolomics experiments that were already successfully finished but focus on improving subcellular and spatial resolution. Protein localization with antibodies is to be carried out.

Causes of Carryover

Expenses for metabolomics and antibody generation have to be spent in batches (multiple samples at once), so exact planning is not possible.
No travel due to travel restrictions.
