Outline of Final Research Achievements |
In the present study, we investigated the impact of perampanel (PER), an anti-epileptic drug targeting AMPA receptors, on β amyloid proteins (Aβ). Aβ is a pivotal pathogenic protein of Alzheimer's disease(AD). We hypothesize that PER might reduce Aβ accumulation by suppressing neuronal hyperexcitability. Using AD mice and cellular models, we demonstrated that acute PER administration suppressed the Aβ production, consequently decreasing soluble Aβ levels in the brain. Moreover, we revealed that chronic PER administration showed a tendency to attenuate Aβ deposition and restore cognitive functions in AD mouse model compared to the control.

アルツハイマー病(AD)は、脳内に異常なタンパク質がたまることで引き起こされる認知症の原因疾患である。AD患者はてんかんとの関連性が高いことが知られているが、AD患者にどのタイミングでどの抗てんかん薬を選択すべきかはコンセンサスがまだ得られていない。本研究は、すでに市場に上梓されている抗てんかん薬ペランパネル(PER)がADの原因タンパク質の一つであるβアミロイド(Aβ)の産生と蓄積を抑制することをマウスと細胞モデルを用いて示した。これは、ADの新たな治療戦略としての可能性を示唆する重要な発見であるといえる。
