| Research Abstract |
昨年度までに、上皮細胞の極性化と脱極性化におけるPAR-aPKC系の具体的な分子機構として、新たにaPKCに直接結合してそのキナーゼ活性を抑制することにより、アピカル膜ドメイン形成を負に制御しているKIBRAを見いだしている。既にaPKCの高発現が胃がんの悪性化に関係している事を報告しているが、今回aPKCとKIBRAとの発現を同一検体群で調べたところ、KIBRAの発現が多いとaPKCの発現が弱くても、胃がんの予後が悪いことが明らかとなった。このことは、この系が実際にヒトがんの悪性化に関わることを支持している。細胞増殖シグナル系とPAR-aPKC 系との具体的な分子機構に関しては、昨年までにLglの新規結合タンパク質としてVprBP及びDDB1を同定した。VprBP-DDB1は、VprBP-DDB1-Cul4A-Roc1ユビキチンE3リガーゼの構成因子であり、このE3リガーゼ複合体の下流でp27及びSkp2の発現制御に関わる事を見いだした。細胞死シグナル系とPAR-aPKC系との具体的な関係とその分子機構に関しては、細胞死誘導因子ASPP2の結合タンパク質として、同定した新規タンパク質が、細胞接着装置と細胞極性因子とを結びつける本態であることを示唆する証拠を得ている。また、他に細胞死誘導タンパク質を同定した。細胞極性と細胞死との関係を明らかにする端緒となる可能性がある。組織幹細胞の自己更新・非対称分裂におけるPAR-aPKC系の具体的な役割に関しては、、FoxG1-Creトランスジェニックマウスを用いたPAR3の神経幹細胞におけるconditionalノックアウトマウスを作成しその解析を進め、PAR3が神経幹細胞の発生初期で重要な役割を果たしていることを示唆する結果を得ると同時に、この過程を制御する新たな経路を見いだした。
