Outline of Final Research Achievements

The following are the main findings of this study with Kuchijirosho-Associated RNAs(KARs):1) Genome sequencing and analysis of the kuchijirosho causative virus identified a novel member of Amnoonviridae. 2) Recombinant KAR proteins (approximately 50 kDa) were expressed in a few strains of Escherichia coli. 3) Intensity and area of ISH signals targeting the KARs were developed mainly in the nuclei at different regions of the brain by the progression of disease in Fugu.4) Little protective efficacy of both a recombinant vaccine and a DNA vaccine against kuchijirosho was observed. These findings will provide an initial step for further understanding the virus infection mechanism and development of a control measure against Kuchijirosho.

Academic Significance and Societal Importance of the Research Achievements

口白症に関する研究は原因ウイルス種が不明なまま停滞していた。しかし,本研究により本症原因ウイルスの全ゲノムの解読に成功したことから,その各分節ゲノムの遺伝子産物を解析・応用することで,ウイルス学的に宿主細胞への感染に直接関わるウイルスレセプターや宿主細胞内でのウイルスの複製機構が解明できるとともに,得られた知見により,ワクチン候補や抗ウイルス薬の標的を特定できることから,口白症のワクチンや治療薬の開発へと活用できる。また,ゲノム情報を用いた本ウイルスの検出技術を駆使して,養殖場でキャリヤーとなっている宿主魚を突き止め,ウイルスの伝播経路を解明することで,感染環の遮断に繋がる知見が得られる。
