Outline of Annual Research Achievements

本研究課題では、HDLコレステロール受容体であるSR-BIの欠損により血漿中で上昇する遊離コレステロールの起源を調査する為に新しいアプローチを試みた。該当年度においてHDL-FC (高密度リポタンパク質中の遊離コレステロール) の放射性標識を評価し、HDL-CE から血中の FC への変換を決定するために、HDL の CE を放射性標識し(辻田)、マウスの血液に投与した(中須賀)。 放射性トレーサーのマウス尾側静脈への投与後 90 分で血漿サンプルを回収した。 本研究成果は、放射性標識 FC が野生型マウスと比較して SR-BI 欠損マウスで実際に増加していることを示しており、CE から FC への変換が血中で起こっているという考えを支持しています。またHDLの主要な構成タンパク質であるApoA-Iの本機構への寄与を評価する目的でApoA-IとSR-BIのダブルノックアウトマウスを作成しました。血漿リポタンパク質はゲルろ過HPLCでの評価(奥平)に加え、密度超遠心法で分離し、ネガティブ染色にて粒子径を測定(高瀬)し、遊離コレステロールを大量に含む大型HDLの出現を確認している。

Reason

SR-BI欠損マウスに加え、apoA-I欠損マウスとのダブルノックアウトマウスの作成に成功している。
これらのマウスのリポタンパク質の組成についてゲル濾過HPLCを用いた分離と比重超遠心法による分離を行い、脂質組成、電子顕微鏡を用いた粒子サイズの観察と画像解析アプリの利用により解析が進んでいる。

Strategy for Future Research Activity

ヒト肝癌由来細胞株HepG2細胞並びにヒト副腎皮質株NCI-H295細胞へSR-BI阻害剤を添加し、輸送活性を遮断し、培養中のHDLの脂質変化を検証する。放射標識したコレステリルエステルを含むHDLを添加することで培養液中において遊離コレステロールが産生されるのか検証する。
