Outline of Final Research Achievements

Na+ and K+ are the major cations in the human body, and the concentration gradient of Na+ and K+ across the plasma membrane plays an important role in secondary active transport and neural activity. On the other hand, knowledge on the function of Na+ and K+ at the endosomal level is scarce. In this study, we used fluorescence bioimaging techniques to show that replacement of NaCl in the culture medium with a membrane-impermeable cation results in rapid transport of endocytosed contents to late endosomes. Surprisingly, the decrease in endosomal pH was suppressed despite transport to the perinuclear region, suggesting that Na+ and its associated ion fluxes regulate the process of endosomal maturation and transport.

Academic Significance and Societal Importance of the Research Achievements

エンドソームは、エンドサイトーシスによって取り込まれた細胞外の物質や細胞膜上の分子を選別し、分解あるいはリサイクルを行うオルガネラである。この過程において、小胞内のH+濃度が徐々に増加することはよく知られており、Na+やK+はH+輸送のための電気化学的勾配の形成、あるいはH+との交換輸送のために濃度が変化するのみと考えられていた。本研究ではNa+がpHに依らず、エンドソーム輸送の指向性を高めることを示唆する結果が得られた。さらに、Na不含培地において、エンドサイトーシス依存的に細胞に侵入するウイルスの感染が抑えられるという予備的な結果を得ている。
