| Outline of Final Research Achievements |
In this study, we show that proteasome dysfunction exacerbates skin damage in the early stages of cutaneous ischemia-reperfusion injury (IRI) using a mouse model that exhibits an aging phenotype. Furthermore, using a cell culture system, we showed that proteasome dysfunction increases oxidative stress by hypoxia/reoxygenation (H/R) and increases reactive oxygen species (ROS) production, but the antioxidant response by activation of the Nrf2 pathway may not function properly. In addition, oxidative stress caused by proteasome dysfunction was effectively suppressed by edaravone (EDA), a free radical scavenger. EDA administration may have a protective effect on skin in the early stages of pressure ulcers in the elderly.
