Outline of Final Research Achievements

In this study, we established to detect an pseudo-tracking of signal transductions during cell state changes by measuring multiple protein informations including signaling molecules. First, we developped a sequential immunostaining (SeqIS) method for complehensive detection of activated signals during cell state changes. Next, we performed SeqIS with 206 antibodies recognize multiple factors including signal transduction molecules for oncogene-induced cellular senescence in a human cell line. The order of signal transduction during cellular senescence was enable to profle by constructing pseudotime by analyzing the SeqIS dataset.

本研究で開発した空間オミクス手法は、細胞状態と空間配置を精緻に捉え、擬似時間解析により特定の細胞状態の将来推定を可能にした。将来的には本技術を疾患組織の解析に応用することで、がんの悪性化や各種疾患につながる特異的な細胞環境やシグナル伝達メカニズムを解明し、新たな疾患の治療戦略確立につながることが期待される。
