Outline of Annual Research Achievements

The aim of this project was to investigate the involvement of TRPV4 in swallowing reflex. So far, TRPV4 expression observed on nerve fibers of the peripheral swallowing-related regions and afferent neurons located in the nodose-petrosal-jugular ganglionic complex (NPJc). TRPV4 majorly localized on large to medium diameter neurons. Topical application of a chemical agonist for TRPV4, GSK1016790A, in the swallowing-related regions dose-dependently facilitated the triggering of swallowing reflex. Prior topical application of an antagonist for TRPV4 significantly reduced the GSK1016790A-induced swallowing reflex. The data obtained in the last year has been published in "Front. Cell. Neurosci." journal. In the coming fiscal year we will conduct research on the involvement of TRPV4 in water-induced swallowing reflex.

2: Research has progressed on the whole more than it was originally planned.

Reason

The research is progressing smoothly as planned. In previous fiscal year, immunohistochemistry studies were conducted to understand the expression
profile of TRPV4 in the SLN-innervated swallowing related-regions and in the nodose-petrosal-jugular ganglionic complex (NPJc). It was also investigated, whether activation of TRPV4 channels by agonists can trigger swallowing reflex, and whether antagonist for TRPV4 can reduce the agonist-induced swallowing reflexes. Some data have already been published in a peer-reviewed international journal. Now we are investigating the involvement of TRPV4 in water-induced swallowing reflex.

Strategy for Future Research Activity

As a future plan various experiments will be conducted to understand the following research questions:
1. Expression profiles of TRPV4 channels in the swallowing-related ganglia in water-deprived rats using immunohistochemistry and qPCR.
2. Whether TRPV4 channels involved in the water-induced swallowing reflex or not.
3. Whether antagonist of TRPV4 channels reduced the water-induced swallowing reflex or not.
4. Whether mechanical stimuli activate TRPV4 channels to trigger swallowing reflex or not.

Causes of Carryover

Need to buy antagonists, antibodies, and reagents.
