Outline of Final Research Achievements |
An architectural transcription factor, high mobility group A2 (Hmga2), is known to be expressed in stem cells and tumor cells. Knock-outing Hmga2 gene causes a pygmy phenotype with bone growth reduction and fat hypo-formation in mice, and it appears consistent with human. Although these strongly suggest that Hmga2 plays an important role in bone development, studies on the molecular pathway are completely unknown. In this study, we focused on the mechanism of osteoblast differentiation by Hmga2, and comprehensively analyzed the functions of Hmga2 using next-generation sequencing technologies such as RNA-seq and ChIP-seq.

Academic Significance and Societal Importance of the Research Achievements |
超高齢社会に突入した現在は骨代謝不調が原因となりQOLが低下するケースが急激に増加しており、骨形成制御機構を解明することは、骨折治癒促進や骨再生療法確立に貢献するとともに、骨粗鬆症等の代謝性骨疾患の治療法開発にも極めて重要である。本研究により、Hmga2依存性の骨芽細胞分化関連遺伝子発現制御機構の存在が示唆された。
