Outline of Annual Research Achievements |
This year, several cuprous-ion-responsive protein labeling reagents (CuRs) have been synthesized, and the chemical structure has been optimized. The cuprous ions-responsive reactions of the optimal CuR have also been studied and validated by HPLC analysis. Moreover, the capability of CuR to label and profile proteins in response to cuprous ions in multiple living cell systems has also been confirmed by western blotting, imaging, and proteomics analysis.

Reason
The optimal cuprous-ion-responsive protein labeling reagent (CuR) has been successfully developed. The labeling mechanism of CuR has been elucidated. CuR has also been successfully applied to multiple living cell systems. Therefore, the research has progressed on the whole more than it was initially planned.

Strategy for Future Research Activity |
In 2023, I will continue to collect data for the publication of the research results obtained. In particular, I will apply the developed optimal CuR to the ATP7A knock-out cells to map the proteins associated with impaired accumulation and export of copper ions by mass spectrometry to study the regulation of copper ions in living systems with genetic misregulation. After this, I will synthesize ligand-tethered cuprous ion-responsive protein labeling reagent (LT-CuR) targeting AMPA receptors to study copper release in the synapse.
