Outline of Annual Research Achievements

(1)我々が開発した変異原性試験の妥当性を検定
変異原性化学物質をハイスループットに検出するバイオアッセイを、ニワトリDT40細胞を使って創った。開発した試験の妥当性を以下のように検証した。大学院生を2014年度から1.5年間米国NIHに派遣し、本バイオアッセイを使い米国National Toxicology Program (NTP) の化学物質ライブラリー(約10,000種類、10Kライブラリー)を解析した。感度および特異性ともに高いことが示された。10Kライブラリーのなかで、これまで変異原性が知られていなかった化学物質の変異原性を新たに検出できた。
(2)変異原性化学物質の作用機序を包括的に解明するシステムの開発
社会のニーズは、化学構造からin silicoに変異原性を予測することにある。in silicoの予測には、質の高い学習データが要る。学習データを構築するには、変異原性化学物質を、その変異誘導の作用機序(=化学物質が作るDNA損傷)毎に分類する必要がある。我々の研究目的は、変異原性化学物質がどんなDNA損傷を作るかを正確に調べるバイオアッセイを開発することである。細胞はDNA損傷の種類毎に別々の修復酵素を持つ。ゆえに、各修復酵素の欠損細胞をゲノム編集で作れば、それらの欠損細胞群の、化学物質への応答性を調べることにより、変異原性化学物質がどんなDNA損傷を作るかを正確に解明できる。我々は、OECD諸国政府が変異原性化学物質検出に使う標準ヒト細胞株(TK6)から約20種類のDNA修復酵素の各ミュータントを既に創った。
(3)小胞体ストレス誘導化学物質を検出するバイオアッセイの開発
小胞体ストレスをおこす薬剤を高感度に解析するバイオアッセイを創った。米国NIHで解析したところ、このバイオアッセイは、高感度であり、かつ米国企業が創ったバイオアッセイの感度と遜色ないことが解った。
