次世代シーケンサーによる老年病の新規ホルモン作用標的の解明と抗加齢医学への応用

Outline of Annual Research Achievements

本研究は個体と臓器の老化・老年病におけるホルモンの分子作用メカニズムを、次世代シーケンサーを活用して解明し、新しいシグナル経路とそれらのクロストーク、新規標的因子、共役因子、修飾因子とその役割を明らかにし、新しい臨床学問領域、抗加齢医学への応用を目指すものである。
(１)　老化と老年病に関わる新規ホルモン受容体標的因子の同定、解析：老化と老年病に関連する核内受容体標的因子に関して、分子機能解析と生体機能解析を進め、老化と老年病における、あるいは老年病治療、抗加齢の標的としての役割を明らかにした。次世代シーケンサーを活用し、ホルモンシグナルと老化、老年病に関わるAPP、EBAG9、マイクロRNA、長鎖非コードRNAをはじめとする核内受容体標的因子を同定し、そのエピゲノム作用、免疫作用、シグナル伝達等の新しい機能を解明した。
(２) ホルモン受容体新規シグナル経路とそのクロストークの解明と老化、老年病における意義：抗加齢、抗老年病作用に関わるホルモン受容体に対する転写活性、作用機序ならびに共役因子との相互作用、蛋白修飾を明らかにした。ホルモンシグナルを協調して調節するRUNX1、FOXP1、CtBP2をはじめとする転写因子と核内受容体の新しい相互作用とその新しい作用メカニズムを次世代シーケンサーを活用してヒトゲノムレベルで解明した。
(３) 新規ホルモン受容体シグナル経路、標的因子の機能・ゲノム解析と抗加齢医学への応用：ホルモン受容体関連因子の遺伝子改変動物、骨、血管、脳神経系等の各組織における老化ならびに老年病表現型を明らかにした。特に、ホルモン作用を媒介するCOX7RP、GGCXとPXRの遺伝子改変動物が老年病、あるいは老年病抵抗性を呈することを示し、新しいメカニズムを解明し、モデル動物を開発した。
