二酸化炭素を燃料化する光捕集分子-人工補酵素-ギ酸脱水素酵素複合体の設計と創製

Outline of Annual Research Achievements

最終年度はギ酸脱水素酵素の二酸化炭素ーギ酸変換反応に対する触媒活性向上を目的として、これまで進めてきたビオローゲン骨格を有する人工補酵素に加えて2,2’-ビピリジニウム塩骨格を有する人工補酵素の設計・合成を進めた。さらに2,2’-ビピリジニウム塩骨格を有する人工補酵素の還元体とギ酸脱水素酵素との相互作用を酵素反応速度論的に解析し、反応速度パラメータを明らかにした。特に2つのピリジン環をプロピレンで架橋した人工補酵素が有効であることを見出した。最後に光捕集機能を有する水溶性亜鉛ポルフィリン、2,2’-ビピリジニウム塩骨格を有する人工補酵素、ギ酸脱水素酵素からなる反応溶液に可視光を照射することによって二酸化炭素が定常的にギ酸に変換されることがわかった.また2つのピリジン環をエチレンあるいはプロピレンで架橋した人工補酵素を用いることによってギ酸生成量が向上することを明らかにした。
平成23年度から進めてきた当該研究では研究開始からギ酸脱水素酵素の二酸化炭素ーギ酸変換反応に対する触媒活性向上を目的として様々なビピリジニウム塩骨格を有する人工補酵素を設計・合成し、これまでに全く明らかにされていなかったこれら人工補酵素の還元体とギ酸脱水素酵素との相互作用を酵素反応速度論的に解析し、反応速度パラメータが決定できた。さらに最終年度には光捕集機能を有する水溶性亜鉛ポルフィリンとこれら人工補酵素、ギ酸脱水素酵素との複合化により、当初の目的であった可視光駆動型の二酸化炭素-ギ酸変換反応を構築することに成功した。
