不完全変態昆虫(コオロギ)の胚形成における位置情報の形成と細胞移動のメカニズム

Research Abstract

23年度はコオロギの初期胚発生における位置情報に関係する因子を特定することを目指しRNAi解析による位置情報関連遺伝子のスクリーニング、および異所性発現系の構築のための実験を行った。特に、主要なシグナル経路であるNotch,Wnt,BMPシグナル経路等に関わる因子の働きをRNAiにより解析した。GFP発現トランスジェニックコオロギの系統を用いたライブイメージングにより、細胞移動を伴う胚原基形成過程でWntとBMPシグナルが重要な役割を果たしていることを示す結果を得た。初期のシグナル因子の働きで、OtdやCaudalが特定の位置に発現し、さらに下流のギャップ遺伝子の発現を誘導することで胚の領域特異化が起こると考えられる。一方、ショウジョウバエのギャップ遺伝子giantやknirpsのオーソログをトランスクリプトームのデータより見いだし、これらが胚の後部で発現することを示す結果を得た。さらに詳細な発現と機能の解析を行う予定である。異所性発現系についてはUAS/GAL4のシステムの構築には至らなかったが、人工制限酵素ZFNに加えて新たにTALENを用いた遺伝子ノックアウトの系を確立することに成功した。これによりターゲット配列の幅が格段に広がり、異所性発現実験を含めた様々な応用の可能性が広がった。

Reason

GAL4/UASシステムの確立はやや難航しているが、一方で人工制限酵素を用いた遺伝子ノックアウトを効率良く行うことができるようになり、蛍光マーカー遺伝子のノックインや異所性発現実験への応用の可能性が広がったため。

Strategy for Future Research Activity

これまでに明らかになってきた初期胚形成に関与するシグナル経路を起点にその上流と下流を重点的に解明し、位置情報の実体とその解読メカニズムに迫りたい。
一方、異所性発現解析のための部位特異的転写制御領域の獲得や、人工制限酵素を用いた遺伝子ターゲッティングを効率的に進めるためにゲノム配列情報が必須である。このため現在進行中のフタホシコオロギ全ゲノム解析の早期完了を目指す。
さらに、UAS/GAL4システムの導入がうまく行かない場合に備え、人工制限酵素によるゲノム編集技術を応用した解析系の構築を進める。
