Outline of Annual Research Achievements |
情報処理が高度に発達した高次脳領域の中心的ニューロンは、莫大な数の神経入力を、シナプスを介した配線伝達により受け取っている。個々のシナプス伝達の重み(比重)は、入力選択的および標的選択的に異なるように設定され、さらにそれぞれのシナプス伝達効率は活動依存的および状況依存的に変化する。本研究は、シナプス伝達の入力依存性、標的依存性、状況依存性の各階層に焦点を当て、その制御機構と機能的意義を分子形態学的視点から解明することが目的である。平成28年度は、線条体中型有棘細胞におけるAMPA受容体の発現制御を中心に解析を行なった。 線条体は大脳皮質や視床からの興奮性入力を統合し、直接路と間接路の相反的な出力経路を通して運動や認知機能を制御している。直接路と間接路を構成する中型有棘細胞において大脳皮質から投射するVGluT1陽性シナプスと視床から投射するVGluT2陽性シナプスの合計4種類のシナプスにおけるAMPA受容体の発現密度を高感度包埋後免疫電顕法で解析した。その結果、いずれのシナプスにおいてもほぼ同じレベルの発現密度であることが判明した。この観察結果は、グルタミン酸伝達において少数の強いシナプスと多数の弱いシナプスからなる小脳、海馬、大脳皮質のシナプス構成とは異なり、線条体では一様な強さのシナプスから成り立っているという特徴を示している。これは、線条体におけるアセチルコリンやドパミンを介した神経修飾が効率よく出力に反映させるための分子解剖学的基盤と考えられる。
