Outline of Annual Research Achievements

1. 副刺激によるT細胞活性化制御：①CD28を介する副刺激はCARMA1-Bcl10-Malt1複合体を介してNFκBを活性化させる。CD28クラスター形成とともに、CARMA1クラスターの形成がNFκB活性化に重要である。CARMA1のGuk領域とSH3領域が会合することを見いだし、この会合はCARMA1の凝集を抑制するが、T細胞活性化によってGuk-SH3会合は、分子内会合から分子間会合となり、CARMA1クラスターが形成されることが判明した。Gukに会合しない変異SH3を発現させたT細胞では、CARMA1欠損T細胞と同様に、T細胞活性化が誘導されなかった。②CD28副刺激にアクチン脱キャッピングタンパク質Rltprが重要であることを見出した。TCR-CD28活性化に伴ってCD28が免疫シナプスのcSMACに集まり、PKCθ－CARMA1をリクルートすることを以前明らかにしたが、RltprはこのCD28とPKCθ-CARMA1複合体を結びつける役割をすると考えられた。
2. 自然免疫シグナルによるT細胞活性化制御：T細胞に発現するTLRによってT細胞に副刺激が誘導されることを見いだし、PAMPに一つとして核酸による活性化を調べているなかで、T細胞は自然免疫系と異なり、CpGでないDNAでも取り込んで活性化されることが解った。核酸はLL37やヒストンなどと複合体を形成してT細胞を活性化する。生体内では死細胞由来のDNAによって活性化されるので、炎症や感染などによって誘導されるDNAに寄って制御されると思われる。DNAによるT細胞活性化は、Th2を強く誘導することを発見した。

Reason

TCRミクロクラスターを介する活性化とその制御機構に関して、副刺激による正負の制御系の解析や、自然免疫シグナルによる制御の解析など、大きく進展した。特に、長年不明であったCD28副刺激クラスターのダイナミックな実態が明らかになり、アクチン細胞骨格との相互制御を明らかにすることでき新たな理解を深めた。T細胞抗原受容体複合体の解析は、もっとも困難な課題で進展の予測の難しい課題であるが、それぞれに分子鎖の全長蛋白の作製の成功、ダイマー構造の精製と一部結晶化の成功、と進展してきている。

Strategy for Future Research Activity

TCRミクロクラスターを介したT細胞活性化シグナルの制御を、正負の副刺激シグナルからの解析や、自然免疫シグナルとの関連の解析など大きく進展したので、より生理的条件 in vivoでのT細胞活性化のイメージング解析を成功させる。昨年までに使用してきたCaインジケーターでは in vivo問題が解決せず、新しいプローブで in vivo imagingに成功させるつもりである。また、同時に計画にある、自己認識シグナルの解析として、自己ペプチドを認識して制御するシグナル解析を行う。さらにTCRミクロクラスターからの活性化によって、T細胞活性化に至る種々のTCR下流シグナル経路を解析してゆく。その一つとして、Ras活性化におけるRas-GRP1などの解析を行なう。
