| Outline of Annual Research Achievements |
膵β細胞の糖・脂質代謝はβ細胞機能に極めて重要な役割を担っている。しかし、①インスリンの分泌制御、②膵β細胞の分化・再生、③糖尿病の発症や病態において、どのような糖・脂質代謝シグナルがそれらのメカニズムの鍵となっているかは殆ど不明である。本研究では、特性の異なる膵β細胞株、膵島、病態モデル動物やヒトのサンプルを用いて包括的メタボローム解析を行い、上記のメカニズムの鍵となる代謝シグナルを解明する。平成28年度は下記の項目について研究を行なった。
１．インスリン分泌における糖・脂質代謝シグナルの同定とその役割の解明：2型糖尿病モデルラットの膵島を用いたメタボロームおよびトランスクリプトーム解析によりインクレチン応答性インスリン分泌(IIIS)の障害に関与する代謝経路を特定した。ゲノム編集技術により小胞型グルタミン酸トランスポーターVGLUT1, -2, -3のトリプルノックアウト膵β細胞株を樹立し、この細胞株でIIISが障害されることを明らかにした。インシリコ類似検索と構造活性相関をもとにしたアプローチによりインスリン分泌促進作用を有する新規低分子化合物を同定し、メタボロミクスによる薬物動態の解析から従来のSU薬と異なる特性を有することを明らかにした。
２．膵β細胞分化・再生における糖・脂質代謝シグナルの同定とその役割の解明：膵β細胞が増殖する時期における単離膵島を用いたメタボローム解析により未分化細胞様の代謝変化を来すことを明らかにした。
３．糖・脂質代謝シグナルに由来する新たな糖尿病の病態マーカーと治療標的の同定： 2型糖尿病モデルラットの組織サンプルを用いた経時的メタボローム解析により糖尿病バイオマーカー候補の責任臓器を特定した。ヒトサンプルを用いたメタボーム解析により、正常、境界型、糖尿病の鑑別や糖尿病腎症のステージの鑑別に有用なバイオマーカー候補を同定した。
