Outline of Annual Research Achievements |
During 26年 we accomplished collection of blood samples by dispatching specialists to the affected after Chernobyl fallout areas, the development of database, and performed DNA extraction from all collected blood samples. We finished biostatistical analysis of obtained data of DNA-exome sequence (point mutations) and mRNA from whole genome sequencing data (expressed rearrangements and point mutations). We discovered a novel chimeric gene, which was a result of balanced chromosomal translocation t(3;10) (q26.32;q11.21). The 5’ portion of the gene was presented by the first 9 exons of TBL1XR containing a LisH region, and the 3’portion was the tyrosine kinase domain of RET. The breakpoint in the RET gene on the mRNA level localized in the same place as in RET/PTC1 and RET/PTC3 in exon 12 of RET. We also found the identical TBL1XR1/RET rearrangement in 1 case among 34 (2.9%) of sporadic pediatric PTCs from Belarus. We also already confirmed that in vitro exposure of normal human thyroid cells (Nthy-ori 3-1) to 1 Gy of γ-radiation resulted in the formation of same rearrangement. The newly found TBL1XR1/RET rearrangement may likely be induced by genotoxic insult, one of which could be ionizing radiation.
