Outline of Annual Research Achievements

本研究は、低線量放射線(LDIR)が生体内微小環境の中で、前がん細胞および間質系幹細胞の遺伝子発現、タンパク発現とその調節機構にどのような影響を与え、発がんに関与するのかを明らかにすることを目的とした。最終年度では、LDIR照射後のmicroRNA(miRNA)の変化について検討した。ヒト不死化細胞 (EBV-B; EBウイスル感染により増殖能を獲得したヒトB細胞株)を正常ヒト骨髄より分離した間質系幹細胞 (mesenchymal stem cell; MSC)との共培養、あるいは単独培養下でLDIR(100mGy)を照射し、LDIRがEBV-B 細胞のmiRNA発現に与える影響を調べた。また、miRNA発現と遺伝子発現変化との関連性およびそのターゲットとなるタンパク発現について検討を行った。その結果、LDIR照射24時間後に単独培養条件下のEBV-B細胞でlet7a, miR-15b, miR-16, miR-21 および脂質代謝関連miRNAのhub-miRNAであるmiR-23bの上昇が認められ、MSC共培養下のEBV-B細胞ではこれらのmiRNA発現の低下が認められた。さらに、これらのmiRNAの共通ターゲットである脂質合成酵素glycerol-3-phosphate acyltransferase(GPAM)の遺伝子(mRNA)およびタンパクレベルの発現変化を認めた(単独培養で低下、共培養で上昇)。 以上より、①LDIRストレスに対してmiRNAを介した遺伝子発現コントロールが機能し、前がん細胞の遺伝子・タンパク発現を変化させること、②生体微小環境は、直接・間接的にLDIRストレス後のmiRNAを介した遺伝子・タンパク発現変化に関与すること、が示唆された。
