Outline of Annual Research Achievements

本研究は、中心体に局在する分裂期キナーゼ群のAurora-A、Polo-like kinase 1（Plk1）の結合蛋白質を探索し、その機能解析を行うことが目的である。これまでの研究で、１）14-3-3ガンマは分裂期特異的にPlk1と結合すること、２）この結合はPlk1-セリン99のリン酸化修飾依存性に引き起こされていること、３）この結合によりPlk1の触媒活性が上昇すること、４）これら一連のPlk1の活性化過程を障害するとスピンドルチェックポイント依存的に分裂中期に停止してしまうこと、５）セリン99のリン酸化修飾は、Akt（キナーゼ）やその上流に位置するPI3キナーゼの触媒活性依存的に引き起こされることを明らかにした。これらの結果は、Plk1の活性調節がこれまで知られているAurora-Aだけでなく、PI3キナーゼ／Akt経路によっても制御されていることを意味する。
