| Research Abstract |
認知機能の低下および認知症の発症、うつ気分の亢進、うつ病の発症は高齢期における心理面の最大の脅威である。その背景として遺伝と環境の要因が想定されているが、両者の関係はそれぞれ個別に検討されてきた。近年、認知機能や感情に影響する遺伝子が同定されてきている。しかし、それらの遺伝子と環境の交互作用に関しては、知見が蓄積されつつあるが、高齢者を対象とした研究は少ないのが現状である。そこで本研究は、これまで認知機能や精神的健康との関連が指摘されている遺伝子を対象に、環境との交互作用を検証することを目的とした。2年間の研究期間を通して、環境要因（幼少期の知能、中年期の仕事、余暇活動）の定量化の作業と遺伝子のタイピングを平行して実施してきた。2年度目は、遺伝子のタイピングに関しては、老化制御や認知機能、精神健康に関わるKlotho遺伝子、Gタンパク質調節因子regulator of G protein signaling（RGS）、およびADARB2遺伝子に注目して分析を進めた。環境要因の評価に関しては初年度に実施した中高年期の仕事内容に加えて、高齢期の余暇活動を、認知的負荷、社会人間関係の量、身体的な負荷、3側面から定量化する方法を開発した。現在、3つの遺伝子に関わるSNPおよび、仕事、余暇活動、小学校の時の学校の成績の自己評価、教育暦を説明変数、そして、現在の精神的健康および認知機能(MOCA)をアウトカム変数として、それらの変数の関連解析を進めている。
現段階では、アウトカム変数に対する遺伝の影響が弱いため、明確な環境要因の交互作用は検出されていないが、一部環境要因を投入することで遺伝の説明率が上昇する傾向も見られる。この結果を受け、研究計画段階では想定していなかったが、調査で用いられている、社会的サポート等の別の環境要因を追加し、研究期間終了後も引き続き分析を続ける予定である。
