Research Abstract |
In this study, we developed a novel cell stretching micro device to observe cellular response to stretching stimuli with high spatial and temporal resolution. First, we have modified our stretching device to reduce the focus drift during the stretching. And we have introduced the fluorescent ratiometric measurement to reduce influence of change in fluorescent intensity arose from the focus drift. Finally, we succeed to conduct observation of cellular response to stretching stimuli with high spatial and temporal resolution. Second, we developed an experimental system to evaluate the intracellular strain field in stretched cell. We combined our cell stretching micro device and the image correlation method to measure the strain in stretched cell. We succeed to evaluated the strain field in the cell during stretch.
