Outline of Annual Research Achievements

本年度は, 分子スケールおよび細胞スケールの統合ナノバイオメカニクスを主な研究課題として推進した. 得られた主要な研究成果は以下のとおりである.
1. クライオ電子線トモグラフィ法を用いて気管繊毛内のモータータンパク質の三次元構造を解析し, ダイニン運動時の構造変化を解明した(Ueno et al., Cytoskeleton, 71, 412, 2014).
2. マラリア, 白血球の免疫機能, 血栓症, がんの血行性転移などにおける微小血管内の細胞流動を解析するための計算モデルを構築した(Takeishi et al., Physiol. Rep., 2, e12037, 2014).
3. 細胞懸濁液の細胞変形やテンソル量を解析するためのGPU計算手法を構築し, 壁面近傍の細胞挙動や振動流中の細胞変形を明らかにした(Nix et al., Phys. Rev. E, 90, 043009, 2014, Matsunaga et al., J. Fluid Mech., 762, 288, 2015).
4. Darkfield internal reflection illuminationシステムを応用したMicrofluidicsの観察手法を開発し, 流路形状とマイクロバブル, 蛍光粒子あるいは蛍光標識細胞を同時観察することを可能にした(Kawano et al., PLoS ONE, 10, e0116925, 2015).
5. マイクロ・ナノバブルを用いて, 酸素過飽和流体を生成できることを示し, マイクロ・ナノバブルを含んだ生理食塩水が血液の酸素化に有効であることを明らかにした(Matsuki et al., Int. J. Nanomed., 9, 4495, 2014).
6. 血液に関連する生命現象プラットフォームの構築に必要となる最先端の実験生体力学および計算生体力学の研究を網羅的に調査し, これらをまとめた総説論文を発表した(Omori et al., Ann. Biomed. Eng., 43, 238, 2015).

Current Status of Research Progress

1: Research has progressed more than it was originally planned.

Reason

研究は当初の計画以上に進展している. 平成26年度は, 分子スケールおよび細胞スケールの統合ナノバイオメカニクスを主な研究課題として推進するだけでなく, 平成27年度以降に実施する予定である細胞懸濁液のテンソル量の解析にも着手している. Microfluidicsの新しい可視化手法やマイクロ・ナノバブルを用いた血液の酸素化手法の開発など当初計画になかった成果も上がっており, 計画を超える速度で研究が進行している. これらの成果は, 雑誌論文16編(うち査読付き14編), 国際会議発表48件(うち招待講演15件)として発表している. さらに, バイオメカニクスの世界大会や計算力学の国際会議など招待講演を多数行うなど国際的に評価されており, 当初の計画以上に進展していると評価している.

Strategy for Future Research Activity

当初計画どおり, 平成27年度は組織スケールおよび臓器スケールの統合ナノバイオメカニクスに関する研究課題を推進する. 特に以下の項目を重点課題とする.
(1) マクロスケールの連続体モデル構築のための, 細胞懸濁液の粒子応力テンソル, 自己拡散テンソル, 粒子拡散テンソルの解析.
(2) 脳動脈瘤, 気管上皮, 皮膚への薬剤輸送などに対する実験と計算の統合的解析
(3) 統合ナノバイオメカニクスに基づくバイオチップの開発
