Role of IP3 Receptor in the Regulation of Synaptic Plasticity, Neuronal Function and Neuronal Development

Outline of Annual Research Achievements

記憶や学習などの脳機能に必要なカルシウムチャネルであるIP3受容体(イノシトール三リン酸受容体)の動作原理とその調節について多くの重要な研究成果を発表した。IP3受容体の2217アミノ酸残基で構成される巨大な細胞質ドメインをIP3存在下・非存在下で結晶化することに成功しX 線結晶構造解析によりIP3依存的なアロステリック構造変化を見い出した。変異体解析でリーフレット構造がこの構造変化を伝達することを解明しIP3によるCa2+放出の仕組みを明らかにした(PNAS,2017)。
既にIP3受容体とアクチンがシナプス可塑性に重要なスパイン形成に関与することを報告しているが、更にアクチンに結合するCaMKIIβが小脳プルキンエ細胞のスパイン形成と伸長を促し、神経発達及び活動依存的なPKCによるCaMKIIβのリン酸化がスパインの過剰発達を抑制することを解明した(PNAS,2017)。シナプスを取り囲むアストロサイト内のIP3Rがシナプス可塑性に関わることを証明した(Glia2017)。
IP3受容体の疑似リガンドであるIRBIT分子(発見して命名済み)はIP3受容体の機能を制御するのみならず、pH調節やシナプス可塑性の調節など多様な機能を示す。しかし異なる標的タンパク質をどのように選択するかが不明であった。IRBITのスプライスバリアントが標的となる標的分子選択性を決める事が明らかになった(PNAS,2017)。
我々はIP3受容体のmRNA が統合失調症の責任因子であるDISC1と結合し長期記憶に関わることを報告し、新たにこのmRNAを樹状突起へ局在化させる因子としてRNG105が必須であることを解明した。RNG105欠損マウスでは、AMPA受容体などのmRNAが樹状突起へ局在化せず長期記憶が著しく低下することを明らかにした(eLife,2017)。
