| Outline of Annual Research Achievements |
先天性好中球減少症（SCN）患者（ELANE変異）の骨髄細胞から作製，樹立した疾患特異的iPS細胞を用いて，以下の結果を得た。①正常者から樹立したiPS細胞，SCN-iPS細胞ともに造血系から骨髄系細胞への分化を確認し，SCN-iPS細胞からの骨髄顆粒球系細胞の増殖，分化障害を確認出来た。この障害は患者骨髄で認められている病態を反映するものであった。②正常iPS細胞を用いた，遺伝子編集からSCN患者に認められた遺伝子変異を導入し，造血系分化，骨随顆粒球系分化を行ったところ，ほぼSCN-iPS細胞と同様な分化障害を作製することができた。分化，成熟細胞が著明に減少し，早期の細胞死が認められたことから，SCNでは細胞死の亢進が，好中球減少の病因のひとつである可能性を示唆していることが明らかとなった。③SCN-iPSの遺伝子編集から，正常遺伝子への修復を試みた。一定の結論を導くことは不可能であったが，一部のクローンでは遺伝子編集されたSCN-iPSが正常に近い骨髄顆粒球系分化を示すものがあり，TALENによる遺伝子編集が，遺伝子修復を可能にすることが示唆された。しかし，すべてのクローンでこの結果を導くことは現段階では出来ておらず，今後，この修復技術の精度の向上と普遍性を明らかとすべき研究を継続している。④同じくELANE変異を有する周期性好中球減少症においても同様の研究を開始しており，同じ遺伝子変異で表現型の違う二つの疾患の病態解析に疾患特異的iPS細胞での造血系分化からの差異を検討している。
