| Research Abstract |
転写因子C/EBPβはBCR-ABL依存性に慢性骨髄性白血病（CML）細胞において発現が亢進しており、CML幹細胞の枯渇と分化を促進することを我々は以前に報告している。本研究課題では、BCR-ABL非依存性に生存していると考えられるCML幹細胞においてC/EBPβの発現を上昇させ、分化・枯渇を誘導することが可能な新規治療戦略への応用を目指して研究を進めている。
当該年度は、まずマウス造血幹細胞株EML細胞を用いて、転写因子C/EBPβの発現をBCR-ABL存在下においても増加させ得るサイトカイン刺激の探索を行った。いくつかのサイトカイン候補の中から、BCR-ABL存在下においてもC/EBPβのmRNAとタンパク両方において増加が認められるものとして、IFNαに着目して以降の研究を進めた。IFNαはSTAT分子の速やかなリン酸化を誘導することから、STATシグナル経路の活性化を介してBCR-ABL存在下にC/EBPβの発現を誘導すると考えられた。
また、C/EBPβ KOマウスとWTマウスの骨髄細胞にBCR-ABLをレトロウイルスによって導入し、これら白血病細胞のIFNα存在下におけるcolony形成能や分化について調べるためにcolony assayを行った。その結果から、IFNαのBCR-ABL発現白血病細胞に対する効果には、C/EBPβ依存性の作用があることが考えられた。現在、CMLマウスモデルにおいてもCML幹細胞に対するIFNαの作用をC/EBPβに着目して研究を行っているところである。

Reason
当初の研究計画通り順調に研究が進められていることから、平成26年度以降に行う予定であった実験についても一部を平成25年度中に進めることができ、それらについても実験結果を得られつつある。C/EBPβの発現をBCR-ABL存在下においても効果的に誘導可能なサイトカインを明らかにすることができ、研究計画に沿って進めた実験によってデータが得られたため、平成25年度はそれらの研究結果をまとめて筆頭演者として米国血液学会にて報告することができた。

| Strategy for Future Research Activity |
in vitroでの実験についてはそのほとんどは進行中であり、実験結果が得られているものがほとんどあるが、CMLマウスモデルでの検討については、予備実験の結果から移植細胞数などモデル作製プロトコールを従来のものから改変する必要性があると考えられたため、現在条件検討を行っている。今後は、順調に進んでいるin vitroの実験系についても行うと共に、in vivoの実験系についてもデータが得られるように重点を置いて進めていきたいと考える。

| Expenditure Plans for the Next FY Research Funding |
アメリカ血液学会参加のための旅費として計上していた旅費について、当該年度は京都大学からの国際学会派遣助成で渡航することができたため、次年度使用分となった。また、実験に用いる消耗品に当てる予定であった物品費についても、研究室に保管されていたものを一部使用できるなど、当初予定したよりも物品購入に当てる費用は少なくなった。
次年度使用となった研究費については、次年度に主にCMLマウスモデルでのIFNαの作用を検討する実験を組んでおり、サイトカイン、FACS用抗体、マウス購入・維持費などの経費が当該年度以上に必要であるため、それらに使用する計画である。
