Outline of Final Research Achievements

Human immunocyte glucan receptor was expressed as a fusion protein in a baculovirus-silkworm expression system and assayed for binding specificity. Recombinant glucan receptor specifically bound to some glucans, but not to other carbohydrates. The glucan binding was inhibited by laminarin, a soluble glucan, and by laminarioligosaccharides, but not by other carbohydrates. Furthermore, the neck domain and oligosaccharide chains of human glucan receptor did not affect the ligand binding activity and specificity of the receptor. These results suggest that recombinant human glucan receptor can be used as a useful probe in identifying ligands in humans and immunostimulating carbohydrates due to its strict binding specificity.
