| Outline of Annual Research Achievements |
生体における肺線維症患者の気管支肺胞洗浄液（BALF）でのプロテアソーム活性や発現を調べた。IIP群が健常群やサルコイドーシス群に比べてChymotrypsin-様活性が高い傾向があった（p=0.089）。新たに20S proteasome活性と呼吸機能との関連を検討したところ、肺活量（VC）実測値および最大吸気量（IC）が正の相関を示したが（r=0.66 およびr=0.82）、一秒率とは負の相関を示した（r=0.72）。比較的経度の患者のプロテアソーム活性が高いことがわかった。喫煙刺激に対しプロテアソーム機能の抑制がどのような影響を及ぼすかを調べるため、C57BL/6の野生型マウス(WT)もしくはプロテアソーム活性が減弱したトランスジェニックマウス（Tg）に5％、60分間の喫煙曝露を週5日間で12週間行った。喫煙曝露を行ったTgマウスはWTと比較し有意に気腔が拡大しており、プロテアソーム抑制は喫煙に対する肺胞破壊が増強することが示された。TgマウスではWTと比較し有意にアポトーシス細胞の増加がみられた。動物種別や採取した部位に関わらず、タバコ煙刺激後のfibroblastはプロテアソーム阻害薬を加えることによってannexin V陽性細胞が増加し、ERストレスも増加した。アポトーシスはcaspase非依存性経路で起こっていると考えられた。8週令のβ5t-Tgマウスおよび同腹のWild typeマウスに0.05 U のブレオマイシンを経気管支投与した。β5t-TgマウスではMasson- trichrome染色の染まりが弱くみられた。また、肺組織におけるTotal soluble collagenはβ5t-Tgマウスでは21日後にはブレオマイシン投与前の値と同じであったが、同腹のWTは高値を示した。Collagen 3 mRNA発現はWTマウスが高い傾向がみられたが、β5t-Tgマウスは経気管投与による死亡回避が難しく統計的に有意な検討は困難であった。
