Outline of Annual Research Achievements |
本研究では、過酸化チタンナノ粒子による放射線増感効果の有効性を明らかにし、これまでにない放射線増感療法を開発し、膵臓癌などの難治癌に対する新たな治療戦略を提案することである。具体的には、Cell-free system(無機物質:ナノ粒子のみの環境)でのヒドロキシラジカル等のReactive Oxygen Species(ROS)生成メカニズムが過酸化チタンナノ粒子の表面構造に大きく依存することが判明した。細胞実験(有機物質環境下)ではγH2AXで確認できるDNA fociの発生細胞数が、過酸化チタンナノ粒子を作用させた後で放射線照射をした群で有意に上昇していた。また、in vivo 環境でも放射線増感が確認された。チタン酸化物ナノ粒子の毒性評価は採血や実験動物の体重の変化から判定したが、顕著な毒性は確認できなかった。過酸化チタンナノ粒子の尾静脈からの投与では腫瘍と肝臓により効率に集積する実験結果を得ており、今後の研究に重要な知見と考えられた。
