Outline of Annual Research Achievements

課題1では,内在性のシトクロムの酸化還元状態を,ラマンイメージングにより無標識に検出できることを示した.既存のATP産生アッセイ法や膜電位計測法と同等のミトコンドリア活性定量感度であることを実証した.さらに研究を展開して,ハイブリッド蛍光ラマン顕微鏡法による化学的状態分析(ラマン分光)と生理状態定量(蛍光標識法)の正確な相関分析を実現する新たな測定法を開発した.蛍光信号とラマン信号の信号混交を抑止する蛍光/ラマン信号の定量かつ客観的分離法を開発した.高速波長変調レーザーと同期した周期変調測定により,これまで定量分析が難しかった細胞種においてもラマン分光による定量化学分析が可能であることを実証した.
課題2では,膜リセプターの光操作を更に展開して,細胞間張力を光制御する新たな技術を開発した.光照射により切断されるタンパク質PhoClを,細胞接着タンパク質E-Cadherinの細胞内ドメインに挿入した融合タンパク質PC-Cadherinを作成した.PC-Cadherinを導入した上皮細胞において青色光照射によりPC-Cadherinが切断され,断片が解離することを実証した.またPC-cadherinや下流分子Vinculinの局在観察により細胞間張力伝達が低下したことを解明した.次に細胞シート内のPC-cadherin発現細胞に光を照射したところ,張力伝達解消に伴う細胞形態変化が観察された.
課題3では,GPCR活性を直接光操作する技術の発展研究として,GPCRの細胞膜とサイトゾルのリサイクリングプロセスを光操作する技術開発を開発した.GPCRの一つであるADRB2について,青色光依存的なタンパク質間相互作用誘起システムCRY2/CIBNを利用し細胞内タンパク質Arrestinとの相互作用を誘導することでエンドサイトーシス,およびリサイクリングが誘起されるを実証した.
