Outline of Annual Research Achievements

H29年度は、より実用的なMembrane-on-Membrane(MoM)シートを開発するため、(1)材料としてのMoMシートの設計ならびに評価、(2)コアユニットであるLYPOzyme開発を目的とする膜場設計について検討した。
(1)MoMシート材料の開発として、脂質膜(脂質二分子膜)が階層的に集密化した材料について開発を行った。これまでにリポソーム固定化ハイドロゲルを作製してきたが、さらに発展させて、リポソーム固定化クライオゲルの調製方法について検討した。本手法では、凍結操作によってリポソームが小さくなり(粒径20nm程度)、pNIPPAMゲル骨格に高密度に固定化できることが明らかとなった。本材料はL-トリプトファンを選択的に吸着する機能を有する。また、ポリマー骨格を必要としない自己組織化ゲルについても検討し、脂肪酸/モノオレイン分子集合体がpH5.0においてゲル化する現象を見出した。本材料の内部では、脂質分子がリポソームの様に秩序高く配列している事から、分子認識能など各種の機能を有すると期待される。
(2)各種の機能を担う脂質膜場の特性解析を行った。エネルギー生産型LIPOzymeとして、クロロフィルを固定化したバイセルにおいて、効率的にJ-aggregateを誘導できることを明らかにした。細胞毒性に及ぼす分子集合体の形態に着目し、集合形態を制御する方法を体系的にデータベース化した。分子認識においては、平衡論的な膜場特性に加えて、外部場印加(例:2.5GHz交流電場刺激)によりキラル認識能が向上することを明らかにした。詳細な膜場特性解析として、膜表層の水和環境に着目した一連の評価手法(誘電分散スペクトル、膜表層結合型蛍光プローブ、表面増強ラマン法)を確立した。

Reason

MoMシート材料に関する基礎的な知見は概ね得られたと考えられる。リポソーム固定化ゲルや脂肪酸ゲルを基盤として、各種の単位操作をマクロスケールで連続的にこなせる材料へと発展させられる。膜場の基礎的な機能評価、ならびに評価手法も概ね確立された。人工光合成(エネルギー生産)、分子認識・キラル変換反応(One-Pot複合型プロセス化学)、革新的なDDSキャリアならびに自己組織化薬剤分子の性能評価のための基礎的な知見も概ね収集できた。

Strategy for Future Research Activity

最終年度となるH30年度において、各種のLIPOzyme ライブラリーに基づいて,機能性膜場を複合化したストレス応答型MoM シートを開発する.
● Case Study ①:多段物質生産プロセスシート:エネルギー生産LIPOzyme(人工光合成の素反応をカスケードさせる);タンパク質合成LIPOzyme(無細胞遺伝子発現系による膜タンパク質を膜場ユニットに直接合成);LIPOzyme シートを重層化し,片面から反応基質が浸透し,反対から生産物が生成するプロセス生産型シートについて、それぞれ作製する.
● Case Study ②:組織賦活化LIPOzyme シート(次世代型人工臓器):(a) 正常細胞に対して高い適合性,(b) ガン性細胞に対して高い毒性,(c) ストレス負荷細胞を賦活化する能力,(d) 抗ストレス機能を兼備したLIPOzyme をカクテル化したLIPOzyme シートを作製し,動物細胞組織あるいは動物細胞に対する毒性を評価する.最終的に既存のポリマー材料シートとの比較を行い,LIPOzymeシートの有効性を示す.上記の知見に基づいて,LIPOzyme シートの基盤・基礎・応用に関する知見を総括し,それらを調製し活用するための方法論を体系化する。
