生殖細胞の発生分化を司るRNA制御機構

Outline of Annual Research Achievements

始原生殖細胞(PGC)の分化に必須な分子Nanos3の下流イベントを解析する目的で、野生型のES細胞とNanso3欠損ES細胞を用いて、始原生殖細胞をin vitro系で分化誘導した。その結果、in vivoの解析同様にNanos3欠損細胞から誘導された始原生殖細胞はPGCの性質を維持できずに次第に消滅していった。その間にどのような遺伝子が変動するかを解析するため、Blimp1-venusでPGCをFACSで分離し、RNAを調整してRNA-seq解析を行った。その結果、発現変動する遺伝子をいくつか同定した。その中で特に興味深い遺伝子に関しては、Cas9系を用いたKO解析を行う予定である。
一方、Nanos3の生後の精子形成における機能を解析するために、条件的KOマウス、及び強制発現マウスを作製して、解析を開始した。
またNanos2とNanos3の機能的な違いの分子基盤を明らかにする目的でNanos2とNanos3のキメラ分子を発現するマウスを作製した。N-末をNanos3に変更したNanos2は胎児精巣で機能しないことがわかった。現在N-末をNanos2に変更したNanos3を作製中である。
DazL-cKO, DND1-cKOを可能にするGS細胞を確立した。

Reason

次世代シーケンサーの不具合があり、RNA-seq解析が遅れたが、繰越を行って解析は完了した。Nanos3のcKOの解析が順調に進んでおり、Nanos2とNanos3の機能的な差がみえてきている。

Strategy for Future Research Activity

ES細胞をもちいた誘導系はやはり、細胞数が少ないことやNanso3の発現が低いことから標的遺伝子の解析など、生化学的な解析は困難である。そこでES細胞を用いて最近我々同定したNanosのパートナー遺伝子DND1を利用して、Nanos2-DND1, Nanos3-DND1をESあるいはGS細胞に発現させて、遺伝子変動や、標的遺伝子の解析に利用する。
