Outline of Annual Research Achievements

本研究では、脳梗塞に特異的な病態のみに効果を発現する虚血応答性核酸医薬を開発し、現在まで有効な治療法が開発出来ていない脳梗塞に対する神経保護両方を実現させることを目的としている。
脳梗塞部位ではpHが6.0-6.5程度に低下するが、周辺に存在する神経保護療法の治療標的部位であるペナンブラ領域はpH6.5-6.8程度であることが判明した。当初開発した虚血応答性核酸は、pH 6.2程度に低下した細胞環境下において標的RNAに対する高い親和性を示すものであったが、フェニルボロン酸部位へのメトキシ基やニトロ基など各種置換基導入により、ボロン酸部位の酸性度(pKa)が制御可能であり、このpKaの制御により分子内ボロン酸エステル形成のon-off制御pHのコントロールも可能である事を明らかとした(JACSに投稿中)。これらの人工核酸への化学修飾技術により、ペナンブラ領域のpHに合わせた虚血応答性核酸の開発に成功した
in vivoでのマウス脳梗塞モデルに対する治療効果における標的mRNAとして中枢神経系の中でも神経細胞や血管内皮細胞に強く発現しているMALAT-1(metastasis associated lung adenocarcinoma transcript 1)遺伝子を選択した。経静脈的に投与したヘテロ核酸はペナンブラ領域の血管内皮細胞や神経細胞に良好に取り込まれ、mRNA抑制効果は脳梗塞部位において80%以上の低下を実現した。一方で、正常動物や脳梗塞と反対側の非虚血部位ではほとんど遺伝子抑制が見られず、明らかな虚血部位選択性を示した。
MALAT-1遺伝子抑制の結果として、梗塞体積が変化したが、その原因としてMALAT-1遺伝子の持つangionegesisや神経保護作用を修飾したことであると考えられた。これらの結果により、虚血病態に選択的に作用する人工核酸により新たな脳梗塞治療が開発出来ると考えられた。
