定量化されたラジカルによるモデル細胞の反応シグナルパス解析

Outline of Annual Research Achievements

従来のように超高真空用チャンバを用いなくてもラジカル源の出口直前での原子状活性種の密度を定量できる活性種照射システムを構築した。構築したシステムにより、今までの我々の研究成果から活性酸素種(ROS)の中で最も殺菌や活性化に関係すると考えられる酸素原子O(3Pj)密度を定量した。このデータに基づき定量化されたラジカル源を用いて、液相中での出芽酵母の活性化や大腸菌の不活性化の効果を検証した。液相中のOHなどの活性種密度をDEMPOなどのトラップ剤を用いて電子スピン共鳴(ESR)装置による測定を試みた。その結果OHが存在し、ラジカル照射時間が長くなるほどESR信号強度が大きくなり、飽和の傾向を持つことが明きらかとなった。一方、OH同士の反応から生成するH2O2の濃度はAmplexRedという試薬を用いて、各種pHの溶液において検量線を立てて定量した。その結果、どのpHの溶液(3ml)においても10分の酸素ラジカル照射(溶液へのO(3Pj)ドーズ量2.4×1018個/cm3)によって生成したH2O2の濃度は10~30μMとなることが分かった。またこのH2O2濃度より少し高い50μMの濃度のH2O2水溶液で大腸菌の殺菌実験を行った。ラジカル照射時と同じ10分間処理してもほとんど大腸菌は殺菌されなかった。これらの結果から、H2O2は殺菌の主要因子ではなく、それ以外のOHかHOOなどのラジカルが殺菌に寄与していることが示唆された。

次年度(平成27年度)に予定していた細胞の活性化・不活性化等のROS系ラジカルフラックス・ドーズ量依存性の評価に関して、既に出芽酵母細胞については完了しているため。

Strategy for Future Research Activity

今後これら液中でのラジカルの密度をESRなどで定量する手法を確立して液中殺菌での主要因となるラジカルの同定を行い、細胞の活性化・不活性化のシグナルパスの解析とメカニズムの解明を行う。また出芽細胞以外の培養細胞としてiPS細胞を予定していたが、もっと基礎的な評価を行うため、iPS細胞の基の細胞としても使われる線維芽細胞を培養細胞を用いて同様な実験をおこなう。またROS系のラジカルだけでなくRNS系、H系のラジカルについても効果を検証する。
