定量化されたラジカルによるモデル細胞の反応シグナルパス解析

Outline of Annual Research Achievements

昨年度構築した原子状活性種の密度を定量できる活性種照射システムを用いて、定量化された基底状態の酸素原子O(3Pj)をリン酸緩衝生理食塩水(PBS)中の出芽酵母と大腸菌に照射した。その結果、昨年度の滅菌水とは異なり、PBS中では1分間の照射で2桁から6桁以上の殺菌が観察され、大腸菌の殺菌効率が高いことが分かった。O(3Pj)をPBSに照射した溶液の残留塩素を定量したところ、1分間の処理で2.5ppm程度となることが分かった。これは大腸菌を次亜塩素酸で十分殺菌できる濃度であり、O(3Pj)照射したPBS中の大腸菌殺菌においては、次亜塩素酸か塩素酸イオンが主要な殺菌因子であることが明らかとなった。
一方、PBS中の出芽酵母においては、30秒程度の照射では、殺菌ではなく10%程度の増殖促進効果があることが確認され、1分間の処理で1桁程度の殺菌効果が観察された。これより、出芽酵母は大腸菌より高い残留塩素耐性があることが確認された。
さらに今年度は、窒素と酸素を導入し、NOx系の活性種を発生させ、出芽酵母の増殖促進・殺菌効果を検証した。NOの密度は紫外分光法により定量し、NOが多い条件では5×10^17cm-3程度の密度となることが確認された。このNOが多い条件下で5分間PBS中の出芽酵母に活性種を照射しても、出芽酵母は全く殺菌されないことが分かった。一方、このNOが多い条件下で、活性種を15秒間照射することで、O(3Pj)照射したときと同等またはそれ以上の増殖促進効果があることも確認された。
また、出芽酵母以外の培養細胞として胎児マウスの線維芽細胞を用いて同様な実験を行った。その結果、出芽酵母と同様に増殖促進の可能性が示唆された。

Strategy for Future Research Activity

今後は、昨年度までに定量評価したデータを基に、酸素系ラジカル(ROS)と酸化窒素系ラジカル(RNS)の相乗効果、これらラジカルとUV・VUV光との相乗効果について評価する。放射光との相乗効果については、大気圧プラズマ源と真空紫外窓により真空紫外光源を作成し、ラジカル源と合わせて使用することで相乗効果を評価する。
この光源の強度が十分に得られない場合は、UV光源などを合わせて利用することも考えて、実施する。
